Derivation and characterization of the iPSC line IITGi001-B from primary human adult dermal fibroblasts

Summary

Adult human dermal fibroblasts purchased from ATCC were reprogrammed using the Y4 combination of episomal plasmids expressing hOCT3/4, short hairpin RNA against p53, hSOX2, hKLF4, hL-MYC, and hLIN28 to generate induced pluripotent stem cells (iPSCs). A representative iPSC clone was extensively characterized to confirm the expression of pluripotency markers, normal karyotype, in vitro trilineage differentiation potential, and genetic identity using DNA fingerprinting. Further, the absence of the integration of episomal plasmids and mycoplasma contamination in the iPSC line was confirmed.
