Selection criteria for clones Patient fibroblasts (OPDM5-KenODWY) were transduced for 24-hours using the Cytotune™- iPS 2.0 Sendai Reprograming kit (ThermoFisher Scientific). Following transduction, Sendai virus was removed, and fibroblasts were maintained in fibroblast medium for 7 days. Transduced cells were then plated on culture dishes seeded with irradiated mouse embryonic fibroblasts (MEFs) in fibroblast medium. On day 8, medium was switched to hiPSC medium (DME/ F12+20%KOSR) and maintained in culture until iPSC colonies formed. At three weeks post- transduction, individual iPSC colonies were mechanically isolated and manually passaged on MEF coated dishes in hiPSC medium twice before they were manually passaged on to one well of a matrigel coated 6-well plate in mTeSR plus. Subsequent passages were performed using 0.5 mM EDTA PBS and E8 medium. Three clones (Cln E, Cln F, and Cln H) were selected for expansion and further characterisation.
