| Notes | Genome-edited Human iPSC Line, SCTi003-A-4, MYH7 R403Q, referred to as SCTi003-A-4, was generated by CRISPR/Cas9 technology, which introduced the c.1208G>A (p.Arg403Gln) mutation into the endogenous myosin heavy chain 7 (MYH7) locus. The R403Q mutation is a well characterized hypertrophic cardiomyopathy (HCM) mutation associated with altered cross-bridge kinetics and hypercontractility, leading to ventricular hypertrophy, myofibrillar disarray, and clinical HCM pathology (Geisterfer-Lowrance et al.; Witjas-Paalberends et al.).
This product’s parental human induced pluripotent stem cell (hiPSC) line, Healthy Control Human iPSC Line, Female, SCTi003-A (Catalog #200-0511) is a well characterized control line derived from peripheral blood mononuclear cells (PBMCs) of a 48-year-old donor. Targeted gene modifications were confirmed by Sanger sequencing. Post-editing, extensive quality control procedures were undertaken in the manufacturing process for SCTi003-A-4 to ensure optimal product performance and reproducibility. SCTi003-A-4 is karyotypically stable, expresses markers of the undifferentiated state, and remains capable of directed differentiation into all three germ layers including cardiomyocyte lineages relevant for cardiovascular research. This genome-edited hiPSC line and its parental hiPSC line provide a robust, isogenic disease modeling tool for studying biophysical mechanisms of HCM, investigating sarcomeric organization, and evaluating therapeutic interventions targeting cardiac contractility. SCTi003-A-4 is manufactured using mTeSR™ Plus (Catalog #100-0276) and is compatible with STEMdiff™ cell culture media, allowing for standardized high-quality maintenance and differentiation to various cell types including functional cardiomyocytes. Cells were obtained using Institutional Review Board (IRB)-approved consent forms and protocols. |
