| Notes | This is an isogenic, ABCA4 gene edited mutant iPSC line derived from the healthy control iPSC line, LVPEIi001-C. It was generated using an exon 44 targeting sgRNA and an engineered en1FnCas9 encoded CRISPR construct. It carries a 12-base pair deletion within the exon 44 of both the alleles of ABCA4 gene. This homozygous genomic edit was confirmed by target-region specific PCR amplification and Sanger sequencing of the genomic DNA. The in-frame deletion resulted in the loss of 4 amino acids within the second nucleotide-binding domain 2 (NBD2) of ABCA4, which causes structural defects that could impair the protein function. The isogenic line was clonally expanded beyond passage 20. This clonal line maintained the expression of major stemness markers (OCT4, SOX2, NANOG, KLF4, SSEA4), demonstrated the ability to differentiate into all three germ layers and exhibited a normal female karyotype. |
