Notes | This was an isogenic, homozygous iPSC knockout line (LVPEIi001-C-1), generated using a high-fidelity engineered en1FnCas9 vector. The line carried a 12-base pair deletion in exon 44 of the ABCA4 gene, which was confirmed through genotyping. The 12-base pair deletion resulted in structural defects in the second nucleotide-binding domain (NBD2) of the ABCA4 protein, impairing its function. The isogenic line was clonally expanded beyond passage 20 and maintained expression of major stemness markers (OCT4, SOX2, NANOG, KLF4, and SSEA4). It also demonstrated the ability to differentiate into all three germ layers and exhibited a normal karyotype. |
