Notes | This is an isogenic, B2M and CIITA gene edited and double knockout iPSC line derived from the healthy control iPSC line, LVPEIi001-C. It was generated using B2M-Exon 2 targeting sgRNA, CIITA-Exon 1 targeting sgRNA and an engineered en1FnCas9 encoded CRISPR construct. It carries compound heterozygous biallelic edits within the exon 2 of human B2M gene and exon 1 of human CIITA gene. The genomic edit in each of the alleles was confirmed by target-region specific PCR amplification and high throughput NGS analysis. The edits caused frameshift in both the alleles and resulted in the total loss of B2M and CIITA functional protein expression. The isogenic line was clonally expanded beyond passage 20. This clonal line maintained the expression of major stemness markers (OCT4, SOX2, NANOG, KLF4, SSEA4), demonstrated the ability to differentiate into all three germ layers and exhibited a normal female karyotype. |
