Notes This is a GMP grade, autosomal recessive retinal dystrophy (ARRD) patient-specific, induced pluripotent stem cell line, carrying a homozygous point mutation within exon 44 of ABCA4 leading to Arg2030Ter. This mutation (c.6088C>T) results in premature truncation of the protein lacking C-terminal nucleotide binding domain. This iPSC line was generated by the reprogramming of donor skin-derived human dermal fibroblasts (HDFs) using GMP compliant processes, xeno-free reagents, cell therapy solutions grade defined culture components and integration-free CTS™ CytoTune™-iPS 2.1 Sendai Reprogramming Kit containing Sendai viral vectors encoding the human OCT4, SOX2, KLF4 and L-Myc genes. This iPSC line maintained proper colony morphology, expressed the stemness and pluripotency markers OCT4, SOX2, NANOG, KLF4, SSEA4, and exhibited a normal karyotype at passage 15.
