Selection criteria for clones Immunofluorescence (IF) analysis confirmed absence of residual Sendai virus proteins and pluripotency with robust expression of NANOG, TRA-1-60, and SSEA4. Trilineage differentiation potential was validated by differentiating into embryoid bodies (EBs) and assessing ectodermal (TUJ1), mesodermal (SMA), and endodermal (β-Catenin) marker expression. Clear upregulation of all three germ layer markers relative to parental iPSCs confirmed the capacity of the clone for spontaneous trilineage differentiation. TaqMan® hPSC Scorecard analysis (ThermoFisher, performed externally) was conducted and their derived embryoid bodies (EBs) to quantitatively assess pluripotency and differentiation capacity. Pluripotency was confirmed by robust expression of core pluripotency genes at levels comparable to established iPSC and ESC standards (n = 13 lines). Differentiation potential was validated by the overexpression of lineage-specific genes in EBs corresponding to all three embryonic germ layers relative to their expression in the parental iPSCs, demonstrating faithful trilineage differentiation. Genomic integrity was assessed by high-density SNP genotyping (Illumina).
